利用近缘种中已发表的微卫星DNA标记引物,对赤点石斑鱼(Epinephelus akeaara)进行跨种PCR扩增,得到12个多态位点,其等位基因数目从2到20不等;表观杂合度范围是0.22240.861,平均为0.595;期望杂合度范围是0.20040.932,平均为0.637.数据分析结果表明,在经过邦佛伦尼校正(Bonferronicorrection)后,这些微卫星位点均符合哈迪-温伯格平衡(Hardy-Weinberg equilibrium),并且各位点两两之间不存在显著的连锁不平衡(1inkagedisequilibrium)现象.阂此可以认为,这12个多态性微卫星位点是研究赤点石斑鱼种群遗传结构的良好分子标记.
Amplified microsatellite loci from 12 Epinephelus akaara cross-species were characterized. Observed number of alleles per locus ranged from 2 to 20. The levels of observed and expected heterozygosities for polymorphic loci were from 0.222 to 0.861 and from 0.200 to 0.932, averaging 0.595 and 0.637, respectively. Analyses showed that all these loci conform to the Hardy-Weinberg equilibrium and no significant linkage dis-equilibrium between any pair of loci after Bonferroni correction was found. Therefore, all these polymorphic loci are good molecular markers for population genetic analysis in E. akaara.
