目的初步研究AK078438基因的功能。方法用已制备的AK078438抗体为一抗,对小鼠心、肝、脾、肺、肾组织进行免疫组织化学(Immunochistochemistry)检测各组织中该基因的表达情况。检测到肾脏组织中表达该基因之后,采用逆转录聚合酶链反应(reverse transcription—polymerase chain reaction,RT—PCR)技术,从小鼠肾脏中提取总RNA,并以之为模板,扩增出AK078438基因。构建绿色荧光表达载体,转染内皮细胞,MTT以及Hoechst荧光染色检测该基因对细胞凋亡的影响。结果免疫组织化学结果显示在小鼠的肾脏组织中检测到该基因表达。RT—PCR得到全长为1065bp的AK078438基因,并成功构建了P—EGFP-AK078438质粒,MTT以及Hochest33258荧光染色检测结果表明该基因的过表达有促进内皮细胞凋亡的作用。结论AK基因在肾脏组织中呈阳性表达,并有促进内皮细胞凋亡的作用,为进一步深入研究该基因功能提供了实验基础。
Objective To study the function of AK078438. Methods Prepared AK078438 polyclonal antibodies, which were suitable to detect endogenous AK078438 in mice, were used as a first antibody in immunohistochemistry. The AK078438 gene was amplified from total RNA extracted from mice kidneys. A eukaryotic expression vector, pEGFP-N1- AK078438, was constructed and then transfeeted into endotheliocyte cells. The induction of apoptosis was measured by MTT and Hoechst 33258 staining. Results Immunohistochemistry revealed that AK078438 was only expressed in kidneys. A 1065 bp fragment was obtained by RT-PCR. The recombinant plasmid pEGFP-mAK078438 was confirmed correctly. The overexpression of recombinant plasmid pEGFP-mAK078438, which was evaluated by MTT and observed by Hoeehst staining, resulted in apoptosis in endotheliocyte. Conclusion AK078438 expresses in the kidney and induces apoptosis in endothelioeyte, which provides a foundation for further research.
