011是从烟草茎中分离得到的一株对植物病原真菌有强烈抑制作用的内生细菌,为了明确其发酵液中的抗菌活性成分,并探索活性成分的大孔树脂吸附条件,本试验采用X-5型、AB-8型和CAD-40型3种树脂对其发酵产物进行了吸附与解吸试验,探讨树脂类型、上样量、解吸剂、解吸剂体积分数及pH对吸附与解吸效果的影响.结果表明,在3种树脂中,CAD-40型树脂的吸附率及解吸率均显著高于X-5型和AB-8型树脂,其最佳上样量为树脂质量的15倍.选择70%的乙醇在pH为7时能获得最佳的解吸效果.
Endophytic bacterium 011,isolated from healthy tobacco stem,had strong inhibitory effects on plant pathogenic fungi.In order to clarify its antifungal ingredients and establish adsorption and desorption conditions of bioactive substances,three types of macroporous resins X-5,AB-8 and CAD-40 were used to determine the effects of resin type,sample weight,desorption reagent,desorption reagent volume fraction and pH on the adsorption and desorption of bioactive substances.The results showed that CAD-40 exhibited the best adsorption and desorption capacity to bioactive substances,and the optimum static adsorption conditions were as followed:fermentation broth was loaded with 15 times of resin mass,and 70% ethanol was used as eluent with pH of 7 as eluent.
