砷是自然界广泛存在的环境毒物,它与人类的许多健康问题息息相关。流行病学已经证实砷暴露可以引起皮肤癌、肺癌、膀胱癌。近年来,应用恶性转化的细胞研究砷化物的致癌机制引起了广泛关注;许多文献报道,砷引起的细胞转化是由于干扰了细胞信号通路。该文就细胞恶性转化模型的建立及砷致细胞恶性转化过程中信号通路改变的分子机制进行了综述,为全面认识砷暴露致癌的机制提供了理论依据,并指出了今后的研究方向。
Arsenic is a widespread environmental toxicant that has been associated with numerous human health problems. Epidemiologic studies demonstrate that exposure to arsenite may cause human cancers of the skin,lung and bladder. Recently, the studies using arsenic-induced transformed cells could provide the direct evidence of arsenic-induced cell transformation, becoming a hot topic. Many attempts have been made to explain the role of arsenic and its derivatives in promoting cellular transformation. The studies have demonstrated that arsenic-induced cellular transformation may occur by perturbation of multiple cellular pathways. The changes of cellular pathways and the development of matrix during arsenic-induced cell transformation were summarized in the present paper, it's useful for understanding the mechanisms of arsenic-induced cancer, and the proposal was given in this field in the future.
