目的 比较不同材料包埋的聚羟基乙酸(PGA)作为支架体外构建组织工程软骨的效果。方法 采用卵磷脂、多聚赖氨酸及纤维粘连蛋白分别包埋PGA支架,并与软骨细胞一起体外培养,观察其亲水性的变化、对细胞生物学行为及合成细胞外基质的影响。结果 以纤维粘连蛋白包埋的PGA支架具有良好的亲水性,改善细胞生物学行为,并显著增强细胞合成细胞外基质的能力。结论 以纤维粘连蛋白包埋的PGA作为软骨组织工程技术中的支架材料,具有较大的应用前景。
Objective To compare the effect of polyglycolic acid (PGA) coated with:different materials as a carrier to create new cartilage in vitro. Methods Chondrocytes were cultured in vitro when they were seeded onto PGA scaffolds coated with different materials, lecithin (LEC), poly-1- lysine (PLYS) and fibronectin (FN), respectively. With light microscope, the hydrophilia of scaffolds and the biological behaviors of chondrocytes were observed. After 4 weeks, the specimens were evaluated by histological staining of HE and type Ⅱ collagen immunohistochemieal staining. Results FN significantly improved the hydrophilia of scaffolds, strengthened the biological behaviors of chondrocytes and promoted the extracellular matrices production of chondrocytes. Conclusion The PGA coated with FN can be used as scaffolds in cartilage tissue engineering.
