目的观察细胞质连接蛋白-170(cytoplasmic linker protein-170,CLIP-170)在小鼠气道上皮损伤与修复过程中的表达及分布变化。方法36只小鼠分为:①博莱霉素(BLM)刺激组30只,经气管插管灌注5mg/kg体重BLM复制小鼠气道上皮损伤模型,于实验第1、4、7、14、28天分别处死6只;②正常对照组6只,同等条件下气管插管灌注生理盐水,于实验第7天处死。观察各组小鼠气道上皮的病理变化;免疫组织化学SP法染色观察CLIP-170在小鼠气道上皮损伤修复过程中的表达和分布变化;Western blot检测各组小鼠肺组织CLIP-170蛋白水平的变化;免疫荧光双标记和共聚焦显微镜技术观察CLIP-170与微管蛋白的共定位关系。结果与对照组比较,CLIP-170的表达在气道上皮急性损伤后有明显上升趋势(P〈0.01),随修复进程其表达有所下调,但差异无显著性意义(P〉0.05),在肺纤维化时再次增加(P〈0.01)。CLIP-170定位于气道上皮细胞胞质内,与微管蛋白有共定位关系。结论CLIP-170在气道上皮损伤后表达增强且与微管蛋白共定位,在小鼠肺气道上皮损伤后修复过程中可能起重要作用。
Objective To observe the changes of expression and distribution of CLIP-170 in the process of airway epithelial injury and repair. Methods The mice were treated with bleomycin intratracheally (5 mg/kg body weight). SP immunohistochemical method and Western blot were respectively used to detect the expression and distribution of CLIP-170 in the airway epithelia. By double-labeled immunofluorescence technique and confocal laser scanning microscopy (CLSM) the co-locational relationship between CLIP-170 and β-tubulin was observed. Results The expression of CLIP-170 in lung tissue was increased significantly (P〈0.01) after injury, then reduced indistinctly (P〉0.05) with the development of the repair process and increased again in the whole fibrosis process (P〈0.01). CLIP-170 was located in the cytoplasm of airway epithelia and co-located with β-tubulin. Conclusion The expression of CLIP-170 was increased strikingly and co-located with β-tubulin after the lung injury. CLIP-170 may play an important role in the process of pulmonary injury and repair.
