目的:通过制作维生素A缺乏模型,检测新生鼠枕颈部骨骼转化生长因子-β1(Transforming Growth Factor-β1,TGF-β1)蛋白表达的情况,探讨枕颈部发育中维生素A对TGF-β1的影响。方法:将健康的雌性C57BL/6小鼠30只随机分成2组,即正常对照组(Normal Control Group,N组)和维生素A缺乏组(Vitamin A Deficiency Group,VAD组),每组各15只。N组给予AIN-93M配方饲料喂养,VAD组给予维生素A缺乏饲料(根据AIN-93M配方修改)喂养。造模成功后,于妊娠E21d,取新生鼠枕颈部骨骼,采用免疫组化检测新生胎鼠枕颈部TGF-β1蛋白表达的情况。结果:经过10周的维生素A缺乏饲料喂养,VAD组小鼠出现维生素A缺乏症状,ELISA检测血清维生素A水平明显低于正常对照组(P〈0.05)。免疫组化检测显示,VAD组检测新生胎鼠枕颈部TGF-β1蛋白表达水平降低,与N组比较差异有统计学意义(P〈0.05)。结论:发现维生素A缺乏导致新生胎鼠枕颈部骨骼中TGF-β1蛋白表达降低,可能与维生素A与TGF-β1信号通路的相互作用有关。
Objective:To observe expression on transforming growth factorβ1(TGF-β1)in mice embryo through vitamin A deficiency in pregnant mice model,which aim to investigate the relationship between vitamin A and TGF-β1in development of the craniovertebral junction.Methods:30female C57BL/6mice were divided into 2groups,namely normal control group(N)and vitamin A deficiency group(VAD),15 mice in each group.N group were fed with normal diet,while VAD group were fed with vitamin A deficiency diet.On 21 st gestational day after modeling success,craniovertebral junction of neonatal mice were taken for detecting the protein expression TGF-β1by immunohistochemistry.Results:After 10 weeks of vitamin A deficiency diet,VAD group female mice showed symptoms of vitamin A deficiency,and serum vitamin A level were significantly lower in VAD group than that in N group(P〈0.05).And the protein expression levels of TGF-β1 were significantly lower in VAD group than that in N group(P〈0.05).Conclusion:In our study,we suggest that vitamin A deficiency could decrease the protein expression level of TGF-β1in craniovertebral junction of neonatal mice,which might be related to the interaction between vitamin A and TGF-β1signaling pathway.
