目的:研究山豆根多糖免疫调节作用的机理。方法:分离正常小鼠脾脏淋巴细胞体外培养,在细胞培养体系中分别加入终浓度为50、100、200、400 mg/L山豆根多糖及其配合刀豆素(5 mg/L)或脂多糖(10 mg/L)作用48 h,用MTT法测定细胞的增殖程度。细胞培养体系中分别加入终浓度为50、100、200、400 mg/L山豆根多糖,分别作用12 h、24 h、36 h、48 h采用G riess法测定上清液中NO的含量。结果:山豆根多糖浓度为100 mg/L时显著促进体外培养的小鼠脾脏T、B淋巴细胞增殖;50 mg/L~200 mg/L的山豆根多糖与小鼠脾淋巴细胞作用24 h即可显著促进细胞分泌一氧化氮。结论:山豆根多糖具有促进体外培养的小鼠脾脏淋巴细胞增殖和分泌一氧化氮的作用。
Objective:To elucidate immunomodulatory mechanism of Sophora subprosrate polysaccharide.Methods: The murine splenic lymphocytes cells were incubated in RPMI-1640 tissue culture medium supplemented with ConA(final concentration 5 mg/L) or LPS(final concentration 10 mg/L).Sophora subprosrate polysaccharide was added into the RPMI-1640 tissue culture at concentrations of 0,50,100,200 or 400 mg /L,respectively.The cell proliferation and secretion of nitric oxide were observed at 12 h,24 h,36 h and 48 h after incubation.Results: Sophora subprosrate polysaccharide siginificantly promoted proliferation of T lymphocytes and B lymphocytes at concentrations ranging from 100 mg/L to 400 mg/L(P0.05~0.01).After incubation of 24 h,Sophora subprosrate polysaccharide markedly increased the production of nitric oxide(NO) in murine splenic lymphocytes at concentrations ranging from 50 mg/L ~200 mg/L(P0.05)and the NO level kept a high level in the following 24 hours(P0.01).The NO secreted by splenic lymphocytes treated with Sophora subprosrate polysaccharide at concentration of 400 mg/L rose markedly beginning from 24 hours and kept a high level in the following 24 hours(P0.01).Conclusion: Sophora subprosrate polysaccharide was able to stimulate proliferation of splenic lymphocytes and increase the production of nitric oxide of the treated cells in vitro.
