Objective： To establish the method for determining the contents of total polysaccharides in the ferrmented Gekko Powders. Methods： Total polysaccharides in the ferrmented Gekko Powders were extracted by ultrasonic water extraction. UV-VIS spectrophotometry was adopted to determine the content of total polysaccharides. Results： The polysaccharides contents of the fermented Gekko powders was 13. 4628mg/g,RSD was 4. 19%（ n = 5）. Conclusion： The determination of total polysaccharides in the ferrmented Gekko Powders use the UV-VIS spectrophotometry,the method is simple and accurate. So the method can be used to determine the content of total polysaccharides in the ferrmented Gekko Powders.
