目的:研究转化生长因子受体1(TβR1)与基质金属蛋白酶-2(MMP2)在头颈鳞状细胞癌中的关系及其对肿瘤细胞侵袭转移机制的调控。方法:构建稳定过表达TβR1的UM-SCC-23细胞系,SDS-PAGE电泳和蛋白质印迹法鉴定细胞系构建。利用Real-time PCR和蛋白质印迹法检测MMP2在稳转细胞系中的表达。细胞侵袭试验检测稳转TβR1的UM-SCC-23细胞系侵袭能力。结果:成功构建了稳定表达TβR1的UM-SCC-23,MMP2在TβR1过表达的UM-SCC-23细胞中基因和蛋白水平表达均增高,稳定表达了TβR1的UM-SCC-23细胞侵袭能力增强。结论:TGF-β/TβR1通路能够上调MMP2表达,促进头颈鳞状细胞癌侵袭能力增强。
Objective: To investigate the relationship between transforming growth factor-beta receptor type-1(TβR1) and matrix metallopeptidase 2(MMP2) in head and neck squamous cell carcinoma,and to examine the influence of TβR1 and MMP2 on the regulation of cancer cell invasion.Methods: We generated a stable UM-SCC-23 cell line expressing His-tagged TβR1,clonal cell lines were selected based on the level of TβR1 by SDS-PAGE and Western blotting analysis.Expression of MMP2 was detected by real-time PCR and Western blotting in the stable cell lines.TβR1 overexpression promoted UM-SCC-23 cell invasion by cell invasion assay.Results: Stable transfection UM-SCC-23-TβR1 cell model has been successfully established.MMP2 was significantly increased in stable transfection UM-SCC-23-TβR1 cell lines compared to the control cell lines.UM-SCC-23 cancer cell lines with a stable expression of TβR1 exhibit an increase in cell invasion abilities.Conclusion: This study suggests an additional role of TGF-β/TβR1 and MMP2 in oral squamous cell carcinoma(OSCC).TβR1 upregulate the expression of MMP2 and promote the invasion of OSCC cancer cell.
