目的制备抗人非小细胞肺癌(NSCLC)的单克隆抗体(McAb),并对其生物学特性及特异性进行鉴定。方法以人肺腺癌细胞株SPC-A1为抗原,免疫BALB/c小鼠,常规细胞融合,间接细胞ELISA法筛选阳性细胞克隆;多种细胞间接ELISA及间接免疫荧光法鉴定其特异性;western blot分析其特异性结合抗原的相对分子量;免疫组化染色分析单抗的组织特异性。结果得到1株高效价的、稳定分泌IgG1抗人NSCLC单抗的细胞株(NJ488-1),纯化后效价为2×106;间接细胞ELISA及间接免疫荧光法证实NJ488-1能特异地识别肺癌细胞系,其识别抗原定位于SPC-A1细胞胞浆区;western blot显示NJ488-1特异性结合的抗原相对分子量(Mr)为70 000左右;免疫组化结果表明NJ488-1与NSCLC组织有强阳性反应。结论成功地制备并鉴定了抗人NSCLC单抗NJ488-1,为肺癌的进一步研究及NJ488-1的临床应用奠定了基础。
Objective To prepare the monoclonal antibody(McAb) against human non-small cell lung cancer(NSCLC) and identify its biological characteristics and specificity.Methods BALB/c mice were immunized with human lung adenocarcinoma SPC-A1 cells and the spleen cells of immunized mouse were fused with SP2/0 myeloma cells.The positive clone which produced McAb against SPC-A1 cells was screened by indirect ELISA.The specificity of McAb was identified by indirect ELISA and indirect immunofluorescence assay.The relative molecular weight of the antigen recognized by McAb was measured by western blot.The tissue specificity of McAb was identified with immunohistochemical staining.Results A cell clone which keeped secreting high-titer IgG1-type McAb(NJ488-1) against human NSCLC cells was obtained.The titer of the purified NJ488-1 was 1∶2×106.Indirect ELISA and immunofluorescence assay demonstrated that the NJ488-1 specifically recognize the antigen located in the cytoplasma of SPC-A1 cells.Western blot showed that the relative molecular weight of the antigen recognized by the McAb was about 70 000,and the immunohitochemical staining indicated that the NJ488-1 strongly reacted with human NSCLC tissue but no reaction with human small-cell lung cancer tissues,pulmonary pseudotumor or breast cancer tissues.Conclusion The McAb against human NSCLC cells,named NJ488-1,was successfully prepared,and its main properties were identified.The McAb provided a useful means for basic research and clinical application of lung cancer.
