目的:筛选能够转化白头翁皂苷的真菌,并对该菌株进行分类鉴定。方法:将白头翁皂苷加入培养基中诱导真菌分泌白头翁皂苷糖苷酶,并通过薄层层析技术检测转化产物。再采用经典的形态学特征观察和现代的ITS—rDNA测序分子生物学分类技术相结合对真菌进行鉴定。结果:筛选出能够生物转化白头翁皂苷的RCEF4894菌株,该菌株被鉴定为棘孢曲霉Aspergillus aculeatus Iizuka。结论:棘孢曲霉RCEFd894菌株能够被诱导产生白头翁皂苷糖苷酶,将多糖基的白头翁皂苷水解成低糖基的皂苷。
Objective: The fungi which could biotransform pulchinenoside were selected, and they were classified and identified. Methods: The pulchinenoside was supplemented into the culture medium to induce secretion of pulchinenoside - glycosidase by the fungi, and the pulchinenoside was detected by silica gel thin - layer chroma- tography. The fungi was identified based on its ITS -rDNA sequence and the morphological characteristics. Resuits: The RCEF4894 strain biotransformed pulchinenoside was isolated and identified as Aspergillus aculeatus Iizuka. Conclusion: The RCEF4894 strain could be induced to produce the pulchinenoside -glycosidase, which hydrolyzed the polyglycosylation of pulchinenoside into underglycosylation of pulchinenoside.
