目的研究控制性超排卵(COH)对胚胎植入前小鼠子宫内膜蛋白质表达的影响,探讨体外受精一胚胎移植时子宫内膜容受性的生物学标志物。方法将12只ICR雌性小鼠随机分为实验组和对照组,每组6只。实验组采取促排卵措施,对照组未作任何处理。通过双向电泳和质谱分析技术研究COH对小鼠子宫内膜蛋白表达的影响。结果实验组蛋白电泳图谱与对照组有明显差异,2倍以上差异表达蛋白总数为89个。截取其中16个蛋白质点进行了质谱分析,检测出9种有意义的差异表达蛋白。结论COH可导致子宫内膜多种蛋白质表达改变,通过筛选子宫内膜容受性的生物学标志物,可以为提高COH周期的胚胎种植率和妊娠率提供理论依据。
Objective To investigate the effect of controlled ovarian hyperstimulation(COH) on expression of protein profile in preimplanted endometrium of mice. Methods Twelve ICR mice were randomly divided into experimental and control groups, COH was given to mice in experimental group. Differential expression of proteins in preimplanted endometrium was analyzed by using two-dimensional gel electrophoresis (2D-DIGE) and matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS). Results There were 89 proteins differentially expressed 〉2 fold in the endometrium between experimental group and control group. Among 89 differentially expressed proteins 16 were further examined with MALDI-TOF-MS and 9 significantly expressed proteins were found. Conclusion Controlled ovarian stimulation can lead to changes in expression of protein profile on the preimplanted endometrium of mice, and screening of endometrial receptivity markers may facilitate to raise the implantation and pregnancy rate.
