对籼稻HL—CMS保持系粤泰B种子成熟胚的愈伤组织诱导培养条件进行了初步探讨。结果表明,将存储的成熟种子于50℃处理2d。种子表面清洗消毒后,晾干2-3h,在1800μmol·m^-2·S^-1光照12h/黑暗12h、温度为24~30℃的较大的组织培养室内,采用pH5.9—6.0、添加3.0mg·L^-1 2,4-D的N6培养基上,粤泰B种子的愈伤诱导率可达93%,且愈伤组织的生长状况很好,为不育基因向籼稻HL-CMS保持系粤秦B的遗传转化奠定了基础。
In this paper the inducing and culturing condition of callus for indica rice Yue-tai B were studied. The results showed that under the improved combinative condition the inducible ratio for callus was more than 93%, and the status of the callus growth was very well. The combinative condition included that treating the storage rice seed with 50℃ for 2 days, airing the seeds for 2-3 hours after sterilizing and washing, culturing under 12 h 1 800 μmol·m^-2·s^-1 light/12 h darker, 24- 30℃, pH 5.9-6.0 N6 medium with 3.0 mg·L^-1 2, 4-D. It provided a foundation for the genetic transformation of indica rice HL-CMS maintainer Yue-tai B.
