建立一种同时测定北五味子中五味子醇甲和五味子酯甲含量的高效液相色谱方法。方法:采用超声提取法对北五味子中的五味子醇甲、五味子酯甲进行提取,利用高效液相色谱法测定其含量。色谱条件:Hypersin BDSC8柱(150 mm×4.6 mm,5μm),流动相为甲醇-0.1%醋酸(70∶30),检测波长254 nm,柱温30℃,流速1.0 m L/min,进样量20μL。结果:五味子醇甲在10.00 ng/μL~26.00 ng/μL范围内(r=0.999 4)线性关系良好,五味子酯甲在2.500 ng/μL~12.50 ng/μL范围内(r=0.999 5)线性关系良好;北五味子中五味子醇甲和五味子酯甲的平均回收率分别为100.9%和100.4%,RSD分别为1.11%和1.24%(n=5)。
To develop a rapid method for simultaneously determination of schisandrin and schisantherin A content in Schisandra chinensis(Turcz.) Baill by high performance liquid chromatography(HPLC). ultrasonic extraction for Schisandrin and Schisantherin A in Schisandra chinensis(Turcz.) Baill and then determined by HPLC. The assay was conducted on Hypersin BDSC8 column(150 mm ×4.6 mm, 5μm), methanol-0.1 %ethanoic acid(70 ∶ 30) as the mobile phase, and detective wavelength was 254 nm.The column temperature was30 ℃ and the fiow rate was 1.0 m L/min.The sample size was 20 μL. Schisandrin showed a good linear relationship at the range of 10.00 ng/μL-26.00 ng/μL(r=0.999 4), Schisantherin A showed a good linear relationship at the range of 2.500 ng/μL~12.50 ng/μL(r =0.999 5). The average recoveries of schisandrin and schisantherin A in Schisandra chinensis(Turcz.) Baill were 100.9 % and 100.4 %,the RSD of the method were 1.11 % and 1.24 %,respectively(n=5).
