目的建立UPLC-ESI-MS/MS法同时测定大鼠尿液、粪便和胆汁中原儿茶酸、异荭草素、野黄芩苷、荭草素、牡荆素、木犀草苷和槲皮苷7种物质的分析方法,并对注射用复方荭草(荭蓼、灯盏细辛等)在大鼠体内的排泄规律进行研究。方法生物样本经甲醇处理,采用Waters Acquity BEH C18色谱柱,乙腈-0.1%甲酸水为流动相,电喷雾离子化,多反应离子监测,分别测定大鼠静脉注射注射用复方荭草后排泄物中7种原形成分。结果 7种成分在大鼠尿液中呈良好线性关系,日内、日间精密度和准确度良好。大鼠静注40 mg/kg注射用复方荭草后,分别有4.98%~22.58%的原形成分在尿液中排出,在粪便及胆汁中各原形药物的浓度均低于最低定量限,同时在各排泄物中可检测出相关的代谢产物。结论该方法特异、快速、灵敏,可用于注射用复方荭草在大鼠体内的排泄研究,该药物在大鼠体内有较广泛的代谢途径,原形药物排出量较少。
AIM To establish a UPLC-ESI-MS/MS method for simultaneously determining seven compounds,protocatechuic acid,isoorientin,quercitrin,orientin,viexin,gauteolin and quercentin in urine,bile and feces of rats and to study the excretion characteristics of Compound Hongcao Injection(Polygoni orientalis Herba,Erigerontis Herba,etc.) in rats.METHODS Following a protein precipitation with methanol for plasma samples,the analysis was achieved on Waters Acquity BEH C18 column,and the mobile phase consisted of acetonitrile and water(0.1% formic acid included).Detection was carried out by the multiple reaction monitoring with ESI ionization source of TQD detector.The concentrations of the seven compounds in the excretions were evaluated after being given Compound Hongcao Injection to the rats.RESULTS Good linearity was achieved for the seven compounds,the intra-day and inter-day precision and accuracy were good.Excretions of seven compounds through urine were from 4.98% to 22.58% after being given 40 mg/kg Compound Hongcao Injection to the rats,and the metabolites founded in the excretions through bile and feces were lower than threshold.CONCLUSION The method is selective,accurate and sensitive.The amount of Compound Hongcao Injection recovered from the excretions after intravenous injected to rats is very low.
