穿山龙总皂苷对胶原诱导性关节炎大鼠血清及滑膜组织肿瘤坏死因子α影响的研究

目的探讨穿山龙总皂苷对Ⅱ型胶原诱导性关节炎(CIA)大鼠血清及滑膜组织肿瘤坏死因子α(TNF-α)的影响,探讨其治疗类风湿关节炎(RA)的免疫调节机制。方法选取48只清洁级Wistar大鼠,随机分成4组:正常对照组(A组)、模型组(B组)、雷公藤多苷片组(C组)和穿山龙总皂苷组(D组),每组12只。B、C、D组大鼠均建立CIA大鼠模型,待模型建立成功后,C、D组大鼠分别给予相应的药物灌胃;A、B组大鼠给予等体积0.9%氯化钠注射液灌胃;均连续给药21d,然后处死大鼠取材。应用双抗体夹心酶联免疫吸附(ELISA)法检测4组大鼠血清TNF-α水平,同时应用逆转录联合酶链式反应(RT—PCR)方法测定4组大鼠滑膜组织TNF-α mRNA表达量。结果B组大鼠血清TNF-α水平及滑膜组织TNF-α mRNA表达量均高于A组,差异有统计学意义(P〈0.05);而C、D组大鼠血清TNF-α及滑膜组织TNF-α mRNA表达量均低于B组,差异有统计学意义(P〈0.05);但C、D组大鼠上述指标比较,差异无统计学意义(P〉0.05)。结论穿山龙总皂苷可能通过调节细胞因子TNF-α的水平来发挥其对RA有益的免疫调节作用。

英文摘要:

Objective To study the influence of total saponin from Rhizoma Dioscoreae Nipponicae (TSRDN) on TNF - α in serum and synovial tissues in collagen - induced arthritis rats in order to investigate its immunal regulation in treating rheumatoid arthritis (RA) . Methods 48 clean Wistar rats were randomly divided into four groups: control group (group A), model group (group B), tripterygium group (group C) and TSRDN group (group D ) with each group 12 rats. CIA rat models were established in group B, C and D before the rats in group C and D were given intragastrie administration of related medi- cines. Rats in group A and B were given intragastric administration of 0. 9% Sodium chloride injection with the same volume. All the rats were treated for 21 days before then were killed. ELISA method was used to detect TNF - α in serum, and RT - PCR method was used to detect the expression of TNF - α mRNA in synovial tissues. Results The TNF -α expression in serum and TNF - α mRNA expression in synovial tissues in group B were both significantly higher than those in group A ( P 〈 0.05 ) . The TNF -α expression in serum and TNF -α mRNA expression in synovial tissues in group C and D were both significantly lower than those in group B ( P 〈 0.05 ) . But the difference between group C and group D showed no statistically significant difference ( P 〉 0.05 ) . Conclusion Regulating the levels of eytokines TNF - α may be the possible mechanism of TSRDN treating RA.
