高蛋白日粮对小鼠胰腺活性氧自由基产生的影响

为探讨高蛋白日粮对小鼠胰腺活性氧自由基产生的影响,试验选用健康雄性C57BL/6J小鼠50只,随机均分成5组,分别于饲喂高蛋白日粮前(0 h)和饲喂后0.5,1.0,1.5,2.0 h眼球采血,并屠宰取出胰腺,测定活性氧自由基及相关指标。结果表明:高蛋白日粮显著提高了血和胰腺中活性氧自由基(ROS)水平,其值随蛋白质在体内的消化吸收而变化,且在1.0 h达到最高峰。同时血浆和胰腺中MDA含量也显著提高,且CAT活性、GSH含量和T-AOC都显著降低。这些结果表明,高蛋白日粮能使小鼠胰腺产生ROS,其生成水平随蛋白质的消化吸收而变化,而且氧化负荷导致胰腺抗氧化防御系统能力下降。

英文摘要:

This present work was undertaken to investigate the effect of high protein diet on ROS generation in pancreas of mice. For this purpose, fifty of male C57BL/6J mice were divided into five groups. After having fasted overnight, four groups consumed high protein diet. Then all animals were slaughtered before (0 h) and 0.5, 1.0, 1.5 and 2.0 h after high protein diet intake. Blood and pancreas samples were obtained for measuring ROS and indicators of oxidant and antioxidant. The results show that high protein diet significantly enhanced levels of ROS in blood and pancreas ( P 〈 0.05 ), its level changed with digestion and absorption of protein and its peak effect occurred 1 h after high protein intake. In addition, MDA content was significantly increased and CAT activity, GSH content and T-AOC were significandy decreased in plasma and pancreas of mice. These data demonstrate that high protein diet could induce ROS generation in pancreas and its level changes with digestion and absorption of protein, and oxidative load made antioxidant capability decrease.
