目的深入分析AKR/J小鼠的与胆固醇代谢相关的表型特征及其内在联系,探讨其作为人类疾病模型的价值。方法AKR/J小鼠为实验组,BABL/C小鼠为对照组,取两组小鼠毛发镜下观察;取两组小鼠的肾上腺、卵巢、睾丸,冷冻切片后进行油红O染色;检测并比较两组小鼠肾上腺相关激素(皮质素、醛固酮、ACTH)水平;检测并比较血常规、血糖、血脂(甘油三酯,总胆固醇)、血电解质及血压。结果AKR/J小鼠内部毛发结构排列紊乱,肾上腺脂类含量明显少于对照鼠,皮质素、醛固酮、总胆固醇浓度比对照小鼠有显著性降低(P〈0.01);血液Na+、Cl-、Mg^2+浓度及平均血红蛋白含量低于对照鼠,Ca^2+浓度高于对照鼠(P〈0.05),其他各项指标均无显著性差别。结论AKR/J小鼠是人类毛发结构缺陷、肾上腺皮质机能减退等疾病的天然模型,也是胆结石及动脉粥样硬化的抗性模型。
AKR/J mice have several significant strain characteristics. This paper made an in-depth analysis on the phenotype characteristics of AKR/J mice, and evaluated their applications as human disease models, such as cholesterol metabolism-related diseases. Methods:AKR/J and BAB1/C mice were set as the experiment group and the control group. The hair of mice was observed under microscopy directly; The tissues of adrenal glands, ovaries and testes were dissected for oil red 0 stain after being frozen and sliced; The adrenal gland-related cortical hormones, such as the cortine, aldosterone and ACTH, were measured; Meanwhile, blood routine examination items such as blood glucose, blood lipids (triglyceride and total cholesterol) , blood electrolytes and blood pressure were examined. Result: Adrenocortical lipid depletion and interior hair derangement were verified in AKR/J mice. Compared with the control, the concentration of cortine, aldosterone, total cholesterol in AKR/J mice were significantly decreased (P 〈 0.01 ). The concentration of blood Na + ,Cl- ,Mg^2+ and the average hemoglobin content were lower than those of control mice (p 〈 0.05) ; Other items were no significant difference between the AKR/J mice and the control mice. Conclusion: The AKR/J mice are ideal animal models for human diseases researches such as hair structure defect, adrenal hypofunction, gall-stone and atherosclerosis.
