通过反复回交——筛选目的基因的手段,将C57BL/6-Tg(UBC-GFP)30Scha/J转基因小鼠的绿色荧光蛋白基因(GFP)分别向BABL/C、CBA/J、DBA/2J、AKR/J、FVB/J等品系小鼠导入,得到了5种不同遗传背景且带有GFP基因的导入系小鼠.冰冻组织切片等方法显示GFP导入系小鼠的皮肤、肝、肾、心、肺等均有不同程度的绿色荧光蛋白表达,不同遗传背景对GFP的组织表达谱、血液生理指标及繁殖性能未见影响.本实验采用基因导入法培育了5种新的带有GFP导入系小鼠,这些不同遗传背景的基因导入系小鼠为移植生物学研究提供了材料.
Using the method of repeated backcrossing and screening target gene, the experiment introduced the green fluorescent protein (GFP) gene of C57BL/6-Tg (UBC-GFP) 30Scha/J mouse into five different inbred strains including BABL/C, CBA/J, DBA/2J, AKR/J, and FVB/J respectively, and finally obtained B6. D2-GFP, B6. CB-GFP, B6. FVB-GFP,B6. C-GFP and B6. AK-GFP congenic mice with the GFP gene. Frozen tissue sections showed that GFP was, in different degree, expressed in tissues like skin, liver, kidney, heart and lung, etc. Different genetic background had no effect on the expression profile of GFP, the blood physiological parameter and reproductive performance. Five kinds of congenic mice with GFP bred in this experiment provide donor materials for the researches of transplantation biology.
