目的用离子排斥色谱法检测焦麦芽中可能存在的丙烯酰胺。方法以水提取焦麦芽中的丙烯酰胺,固相萃取柱净化浓缩,离子排斥色谱分离,紫外检测器测定其含量。色谱条件为Shimadzu SCR-102H柱(300 mm×7.8 mm,7μm);流动相为0.01 mol·L^-1H2SO4-乙腈(70:30);检测波长195 nm;流速0.6 ml·min^-1。结果丙烯酰胺0.02-5.00 mg·L^-1与峰面积线性良好(r=0.9996);检出限0.02 mg·L^-1,RSD=3.97%,平均回收率96.0%。结论所建方法简单、可靠,可用于炮制中药材中丙烯酰胺的检出。
OBJECTIVE To establish a method for the determination of acrylamide in coking malt by Ion - exclusion chromatography. METHODS The acrylamide in coking malt was extracted with water. The extract was purified and concentrated with solid phase extraction column, then separated and detected by IEC, respectively. Acrylamide was separated on a column of Shimadzu SCR - 102H (300 mm × 7.8 mm, 7 μm) with 0.01 mol· L^- 1 H2 SO4 - acetonitrile (70 : 30) as mobile phase. The flow rate was 0.6 ml· min ^-1 and the detection wavelength was set at 195 nm. RESULTS The calibration curve was linear between 0.02 mg· L^-1 and 5.00 mg· L^-1, and the correlation coefficient was 0. 9996. The average recovery was 96.0% with RSD of 3.97%. CONCLUSION The method is simple, fast and accurate. It can be used for the investigation of acrylamide in processing of Chinese medicine.
