目的:探索艾灸治疗实验性类风湿性关节炎(RA)大鼠海马中枢的功能机制。方法:将SD大鼠随机分为空白对照组、RA模型组、艾灸组,用弗氏完全佐剂制造实验性RA病理模型,艾灸"肾俞"、"足三里"穴治疗,用免疫组化法测定大鼠海马糖皮质激素受体(GR)表达,用原位杂交法检测海马褪黑激素受体1B(MEL1B)表达。结果:模型组与空白组比较,GR表达有增高趋势(P〉0.05),艾灸组显著低于模型组(P〈0.01)。模型组与空白组比较,MEL1B光密度值显著增高(P〈0.01),艾灸组明显低于模型组(P〈0.05)。结论:艾灸对实验性RA海马中枢有良性调整作用,海马GR、MEL1B在艾灸治疗实验性RA大鼠抗炎效应中具有重要作用。
Objective:To explore the mechanism of moxibustion treating hippocampal function of experimental rheumatoid arthritis(RA)rats.Methods:Sprague-Dawley(SD)rats were randomly divided into normal control group,model group and moxibustion group.RA animal models were made by Freund's Complete Adjuvant(FCA),Shenshu(BL23)and Zusanli(ST36)acupoints were selected to make moxibustion.The hippocampal glucocorticoid receptor(GR)expression was detected by immunohistochemical method,and the hippocampal melatonin receptor 1B(MEL1B)expression by in situ hybridization method.Results:Compared with normal group,the GR expression of model group had an increasing trend(P〈0.05),that of moxibustion group was significantly lower than that of model group(P〈0.01).Compared with control group,MEL1B optical density values of model group increased significantly(P〈0.01),that of moxibustion group was significantly lower than that of model group(P〈0.05).Conclusion:Moxibustion has on good regulation effect on experimental RA hippocampal central functions.GR and MEL1B in hippocampus play an important role in moxibustion treatment of experimental rats with anti-inflammatory.
