目的 分析ANKRD18B基因在大肠癌组织中的甲基化发生情况及对其表达的影响.方法 采用MSP(methglatim specific)方法检测癌旁正常组织和大肠癌组织以及大肠癌肿瘤细胞株ANKRD18B基因甲基化发生情况;采用去甲基化药物5-aza-dC处理大肠癌肿瘤细胞后,采用RT-PCR法检测ANKRD18B基因mRNA的表达水平.结果 癌旁正常组织中的ANKRD18B基因甲基化发生率为0% (0/20),大肠癌组织中ANKRD18B基因甲基化发生率为59% (24/41);去甲基化药物5-aza-dC处理大肠癌肿瘤细胞株后ANKRD18B基因mRNA表达均明显升高.结论 ANKRD 18B基因在大肠癌中均发生甲基化,其甲基化的发生抑制该基因的表达.提示ANKRD18B基因甲基化可能在结直肠发生过程中起到重要的作用。
Objective To analyze ANKRD18B gene methylation and expression in colorectal cancer. Methods Methylation specific PCR was used to detect the DNA methylation of ANKRD18B gene. The RT-PCR was used to analyze the expression of ANKRD18B gene in tumor cells with 5-aza-dC-treated treatment. Results The hypermethylation of ANKRD18B was detected in colorectal cancer tissues (59%, 24/41) and (0%, 0/20) in normal tissues. ANKRD 18B showed hypermethylation and its expression decreased in colon cancer cell line SW480 and HT29. After treatment with demethylation drug, the expression of ANKRD18B gene increased. Conclusion ANKRD18B gene hypermethylated, which inhibited the expression of the gene in colorectal cancer. ANKRD18B gene methylation may play an important role in colorectal cancer.
