目的分析3种一氧化氮合酶(NOS)亚型在结肠癌中的表达差异,探讨神经元型一氧化氮合酶(n NOS)对结肠癌细胞增殖和迁移功能的影响。方法利用流式细胞术、Western blot、免疫荧光、组化等技术,对6株结肠癌细胞(SW480,SW620,RKO,LOVO,HT29,M5)和10例组织样本中NOS的表达进行分析。而后分别以NOS抑制剂干预NOS的功能活性,分析NOS对结肠癌细胞功能的影响。结果 3种NOS在结肠癌细胞和结肠癌组织中均有表达,以胞浆表达为主,部分核表达。流式细胞术和Western blot定量显示n NOS在所有样本中表达最强(分别为239±4.7~693.3±38.0和0.263±0.05~0.696±0.098),诱导型一氧化氮合酶(i NOS)居中(分别为42.7±13.6~236±34.0和0.079±0.04~0.291±0.006),内皮型一氧化氮合酶(e NOS)微弱或不表达(分别为13±7.4~68±13.6和0.019±0.004~0.123±0.004)。增殖和迁移功能实验显示,同时抑制3种NOS亚型或选择性抑制n NOS,可显著下调结肠癌细胞株的增殖和迁移功能(与对照组比较,抑制增殖迁移能力中位百分数为42.5%,P〈0.05),而选择性抑制i NOS效果微弱(仅抑制9.7%中位百分数的增殖迁移能力,P〉0.05)。结论n NOS对结肠癌增殖和迁移起着重要作用,针对n NOS表达干预治疗可能成为将来结肠癌临床治疗的手段之一。
Objective To analyze different expression of the three kinds of NOSs,and thus to evaluate its role in colon cancer.Methods Flow cytometry,Western Blot,immunofluorescence and immunohistochemistry technology were used to analyze NOS expression in six colon cancer cell lines( RKO,LOVO,SW480,SW620,HT29 and M5) and 10 tissue samples.NOS inhibitors were given for cell functional tests.Results NOS were mainly expressed in cytoplasm,while nucleus expression could be detected in certain condition.According to flow cytometry and Western blot,n NOS displayed the highest expression( 239 ± 4.7 ~ 693.3 ± 38 and 0.263 ± 0.05 ~ 0.696 ± 0.098,respectively),i NOS the expression( 42.7 ± 13.6 ~ 236 ± 34.0 and 0.079 ± 0.04 ~ 0.291 ± 0.006,respectively) and e NOS the lowest expression( 13 ± 7.4 ~ 68 ± 13.6 and 0.019 ± 0.004 ~ 0.123 ± 0.004,respectively).Proliferation and invasion functions could be significantly inhibited by n NOS special inhibitor N-PLA and the non-selective NOS inhibitor L-NAME( 42.5%,P〈0.05),but limitedly inhibited by i NOS special inhibitor 1 400 W( 9.7%,P〉 0.05).Conclusion The n NOS might play an important role in the proliferation and invasion of colon cancer,selective inhibition of n NOS might be an effective treatment for colon cancer.
