为了确定蛋白磷酸酶-1(protein phosphatase-1)的催化亚基(PP-1c)在小白鼠不同器官组织(肌肉、卵巢、肾、胃、脾、大脑、心、肝、肺及乳腺)中的表达模式,运用RT-PCR、Western印迹及荧光免疫组织化学技术等实验手段进行了检测和分析.结果表明,在mRNA水平,PP-1c在大脑中表达最高,卵巢及肺中表达次之,在肌肉、肾、心、肝中表达较低,在胃和乳腺中表达最低;在蛋白质水平,肝中表达最高,肾、大脑、肺和乳腺中表达较高,而肌肉、卵巢、心和脾中表达相对较低,胃中表达最低.免疫荧光组织化学实验结果显示,PP-1c的表达也具有明显的组织特异性和细胞特异性.这些结果为进一步探讨PP-1在哺乳动物不同组织器官中的功能提供了重要的实验依据.
To study the differential expression and distribution of the catalytic subunits of protein phosphatase-1(PP-1c) in various tissues of adult mouse,total RNAs and proteins were isolated from the different tissues including muscle,ovary,stomach,kidney,spleen,brain,heart,live,lung and breast.The tissue-specific expression of PP-1c mRNA was analyzed using RT-PCR.The PP-1c protein expression and distribution were examined by Western blot analysis and immunohistochemistry.RT-PCR showed that the mRNAs for PP-1c were expressed at the highest level in brain,the higher levels in ovary,spleen and lung,the low levels in muscle,kidney,heart and liver,and the lowest levels in stomach and breast.Western blot analysis demonstrated somewhat different patterns of protein expression in which the PP-1csubunits were expressed with the highest level in liver,the higher in kidney,brain,lung and breast,the low levels in spleen,heart,muscle,ovary and the lowest level in muscle.Such discrepancy reflects either differential translation or differential stability of PP-1c in various tissues.Immunohistochemistry revealed non-homogeneous distribution of PP-1c in different types of cells from the same tissues,or different levels of PP-1c signals in the tissues.These results provide important information for the further exploration of the functions of PP-1in the major tissues of mammals.
