FUCA1是糖基水解酶家族的成员之一,参与糖蛋白、糖脂等生物活性大分子的分解代谢反应.为了进一步了解α-L-岩藻糖苷酶在鱼类生殖发育中的功能,首次分离了黄鳝FUCA1基因,利用在线软件SMART对所克隆出来的FUCA1基因部分核苷酸序列及其推导的氨基酸序列进行对比分析,其结果表明该序列包含了3个跨膜结构域和4个低拷贝区域.通过RT-PCR检测了FUCA1在黄鳝性腺组织特异性表达状况,FUCA1基因在黄鳝雌性性腺中的表达量高于在雄性性腺的表达量.FUCA1基因在黄鳝雌雄个体间的差异性表达表明,FUCA1基因可能参与黄鳝性逆转生殖发育调控.
The alpha-l-fucosidase1(FUCA1) is a member of the glycosyl hydrolase family and plays an important role in the catabolism of glycoproteins, glycolipids and other bioactive macromolecules reactions. To further explore the functions of FUCA1 in fish reproductive development, the FUCA1 gene has been isolated from ricefield eel for the first time, and comparative analyses of their gene sequences have been conducted by means of online software SMART. It is shown that gene amino acid sequence of the FUCA1 contains three transmembrane domains and four low copy areas. Its specificity of gonadal tissues has also been examined by RTPCR, which reveals that the expression level of FUCA1 in the ovary is higher than that in the testis of ricefield eel. It is therefore claimed that the FUCA1 gene may be involved in the regulation of sex reversal in ricefield eel.
