目的观察衰老对大鼠心肌过氧化物酶体增殖物激活型受体γ(peroxisome proliferator activated receptor γ,PPARγ)表达水平的影响及阿托伐他汀的干预作用。方法选择30只20月龄的Wistar大鼠,随机分为老年组、大剂量阿托伐他汀处理组(大剂量组,10mg·kg^-1·d^-1)和小剂量阿托伐他汀处理组(小剂量组,1mg·kg^-1·d^-1),每组10只;另选10只3月龄Wistar大鼠为青年组。小剂量组和大剂量组大鼠每天给予相应剂量的阿托伐他汀灌胃,共4个月。老年组给予相同体积的生理盐水灌胃,共4个月,青年组未灌胃。采用RT—PCR方法检测大鼠心肌PPARγ mRNA含量,用Western blot方法检测大鼠心肌PPARγ蛋白表达水平。结果与青年组比较,老年组大鼠心肌PPARγ mRNA含量和蛋白表达水平显著降低(P〈0.01);与老年组比较,小剂量组和大剂量组大鼠PPARγ mRNA含量和蛋白表达水平显著增加(P〈0.01),并呈剂量依赖性。结论衰老大鼠心肌PPARγ表达水平显著降低,阿托伐他汀可显著上调老年大鼠心肌PPARγ的表达。
Objective To explore the effects of aging and atorvastatin on the expression of peroxisome proliferator-activated receptor γ(PPARγ)in myocardium of rats. Methods Thirty Wistar rats of 20 months old were divided into three groups:aged group,atorvastatin high dose group(10 mg·kg^-1·d^-1),atorvastatin low dose group(1 mg·kg^-1·d^-1) and other 10 Wistar rats of 3 months old served as young group. Atorvastatin was given by intragastric administration for 4 months. Normal saline was given to rats of aged group by intragastric administration too. The level of PPARγ mRNA was determined by RT-PCR,the expression of PPARγ protein was detected by Western blot. Results The expression levels of PPARγ mRNA and PPARγ protein in aged group were significantly lower than those in young group (P 〈0.01). The expression levels of PPARγ mRNA and PPARγ protein in atorvastatin groups were significantly higher than those in aged group (P 〈0.01). Conclusion Atorvastatin upregulates the expression level of PPARγ in myocardium of aged rats.
