目的研究复方龙垂草水煎剂对四氯化碳(CCl4)致小鼠急性肝损伤的保护作用及其机制初探。方法预给药7 d后,于末次给药后1 h采用腹腔注射的方法注射0.2%CCl4复制小鼠急性肝损伤模型,16 h后取血清检测血清中丙氨酸转移酶(ALT)、谷草转氨酶(AST)活性;取肝脏计算肝指数,检测肝组织中丙二醛(MDA)、超氧化物岐化酶(SOD)指标变化,用Western Blot法检测肝脏组织中Caspase-3蛋白表达情况。结果复方龙垂草水煎剂可降低血清中ALT、AST(均P〈0.01)活性,抑制肝组织中MDA(P〈0.05)上升,上调SOD(P〈0.01),抑制凋亡蛋白Caspase-3表达。结论复方龙垂草水煎剂对CCl4致小鼠急性肝损伤有一定的保护作用,其作用机制与抑制转氨酶及脂质过氧化有关,对CCl4诱导的凋亡信号通路也有抑制作用。
Objective To study the protective effect of Compound Long Chuicao Decoction on mice acute liver injury induced by carbon tetrachloride(CCl4), and to investigate its potential mechanism. Methods All mice except for the normal control group were given Compound Long Chuicao Decoction for 7 days. One hour after last administration,mice were given intraperitoneal injection with 0.2 % CCl4 to induce acute liver injury model. Sixteen hours later,we detected the activities of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in the serum,calculated the liver index,examined hepatic malondialdehyde(MDA)and superoxide dismutase(SOD),and detected the protein expression of caspase-3 in liver tissue by western bolting. Results Compound Long Chuicao Decoction had an effect on reducing the activities of serum ALT and AST(P〈0.01),inhibiting the increase of hepatic MDA(P〈0.05),up-regulating SOD activity(P〈0.01), and suppressing the expression of hepatic caspase-3 in mice with CCl4-induced acute liver injury. Conclusion Compound Long Chuicao Decoction has certain protective effect on CCl4-induced acute liver injury in mice,and the mechanism might be related to its antioxidant activity and inhibition of aminotransferases,and also with the inhibition of apoptosis signaling pathway.
