目的 研究帕金森病(Parkinson’s disease, PD)状态下大鼠丘脑背内侧核(mediodorsal thalamus, MD)神经元电活动的变化。方法 以6-羟多巴胺诱导的PD模型大鼠为实验对象,采用在体玻璃微电极细胞外记录的方法记录大鼠MD神经元的放电频率和放电形式的变化,并与对照组大鼠进行比较分析。结果 PD大鼠MD神经元平均放电频率为(4.66±4.27)Hz,明显高于对照组大鼠(3.32±2.71)Hz;两组大鼠MD神经元放电形式都表现为不规则放电与爆发式放电两种,而PD组大鼠MD内不规则放电的神经元比例高于对照组大鼠(60.78%、49.21%),爆发式放电的神经元比例低于对照组大鼠(39.22%、50.79%),且与对照组相比,PD组大鼠MD神经元放电形式的相关参数均有差异。结论 帕金森病模型大鼠中MD神经元放电频率增高,兴奋性增强,同时放电形式改变,神经元活动的规律异常。
Objective To study the changes of neuronal firing activity in mediodorsal thalamus (MD) nucleus of rats with Parkinson’s disease (PD). Methods The experimental subjects were rats with PD induced by 6-hydrodopamine (6-OHDA). We compared and analyzed the changes of firing frequency and firing forms of MD neurons in PD group and control group with the results obtained with in vivo glass microelectrode extracellular recording method. Results The mean firing frequency of MD neurons in PD group was (4.66±4.27)Hz, significantly higher than that in control group (3.32±2.71)Hz. MD neuronal firing form in the two groups was irregular discharge and explosive discharge, the proportion of neurons with irregular discharge was higher in PD group than in control group (60.78% vs. 49.21%), and the proportion of neurons with explosive discharge was lower in PD group than in control group (39.22% vs. 50.79%). Parameters related to MD neuronal discharge form in PD group were all different from those in control group. Conclusion In MD of PD rats, the neuronal firing frequency increases and excitability enhances. At the same time, firing form changes and neuronal activity is irregular.
