目的 探讨独活寄生汤含药血清对硝普钠诱导的膝软骨细胞凋亡模型增殖的影响。方法 不同浓度硝普钠(SNP)诱导软骨细胞凋亡,MTT法检测软骨细胞半数致死量(LD50)的硝普钠浓度,以确定硝普钠最佳诱导浓度;10%独活寄生汤含药血清干预72 h后,光学显微镜观察细胞形态变化;Hoechst 33258染色,荧光倒置显微镜观察细胞凋亡情况。qPCR法检测各组转化生长因子β1(TGF-β1)mRNA的表达。结果 1 mM硝普钠为最佳诱导浓度;含药血清干预后,软骨细胞的凋亡率降低,干预组低于模型组,空白组软骨细胞中TGF-β1 mRNA的表达最高,模型组最低,干预组介于两组之间,各组比较差异有统计学意义(P〈0.01)。结论 独活寄生汤含药血清能抑制硝普钠诱导体外培养的膝软骨细胞凋亡模型的凋亡。
Objective To investigate the effect of serum containing parasitic soup on the proliferation of knee chondrocytes apoptosis model induced by sodium nitroprusside. Methods Since different concentrations of sodium nitroprusside(SNP) induced chondrocyte apoptosis,the half-lethal dose(LD50) concentration of sodium nitroprusside sodium nitroprusside(SNP) in chondrocyte cell was determined by MTT assay to determine the optimal concentration of sodium nitroprusside(SNP). The morphological changes of the cells were observed by light microscope after treated with 10% for 72 h.After being stained by the dye Hoechst 33258, cell apoptosis was observed by fluorescence inversion microscope. The expression of transforming growth factor-β1(TGF-β1) mRNA was detected by qPCR. Results 1 mM sodium nitroprusside was the best induction concentration. The apoptosis rate of chondrocytes decreased after treatment with serum containing drugs,and the apoptosis rate of the intervention group was lower than that of model group.The expression of TGF-β1 mRNA in the chondrocytes in the blank group was the highest, the lowest in the model group and the level of TGF-β1 mRNA in the treatment group was between the two groups. There was significant difference between the three groups(P〈0.01). Conclusion The serum containing parasitic soup can inhibit the apoptosis of knee chondrocytes apoptosis model induced by sodium nitroprusside in vitro.
