以5,5’-偶氮二水杨酸作为起始剂,异佛尔酮二异氰酸酯作为交联剂,合成了新型的葡聚糖水凝胶作为药物靶向载体,并对载体材料进行了红外、溶胀度及扫描电镜的分析研究。水凝胶的溶胀实验结果说明:随着水凝胶交联程度的降低,其溶胀程度增大,平衡时间延长。扫描电镜观察表面形态验证了充分溶胀的水凝胶具有多孔、均相结构,而且孔径随着交联度的增大而减小。该类水凝胶可作为多肽和蛋白类药物的载体。
A method was developed to prepare novel dextran hydrogels as carriers for colonic drug targeting obtained by copolymerization of dextran with di-(2-hydroxyethyl) 5,5'-azodisalicylate using isophorone diisocyanate as cross-linking agent in dimethyl sulphoxide solution. The properties of the resultant dextran hydrogels were characterized by Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). The results of swelling behavior of hydrogels demonstrated the relation between cross-linking degree and the max swelling ratio. At the same time, the swollen morphology of dextran hydrogels obtained by SEM demonstrated that water channels (porous presented in the SEM photographs) were distributed homogeneously through out the network membranes. The size of the pores intends to decrease with the increase of cross-linking degree. The resultant azo-containing dextran hydrogels can be used as colon specific drug delivery carriers for some bioactive substances.
