目的探讨研究微小RNA(miRNAs)基因区域多态性与非小细胞肺癌易感性的关系。方法选取2012年1月至2014年6月于河北大学附属医院就诊的新发非小细胞肺癌患者50例作为研究组,另选取同期在我院门诊体检中心行身体检查的健康志愿者50名作为对照组。比较2组受试对象血液miRNAs基因区域多态性分布情况,并依据不同临床分期将研究组患者再次分组,比较各小组miRNAs基因区域多态性,分析其与非小细胞肺癌易感性临床关系。结果研究组miRNAs 149rs2292832位点CG和196a2rs11614913位点CC构成比均较对照组显著降低(P〈0.05),而研究组miRNAs 196a2rs11614913位点CT构成比显著高于对照组(P〈0.05);研究组Ⅰ期、Ⅱ期、Ⅲ期和Ⅳ期各样本间149rs2292832位点CG、196a2rs11614913位点CT和CC差异均有统计学意义(P〈0.05),且149rs2292832位点CG、196a2rs11614913位点CC和非小细胞肺癌易感性存在负向相关性(r=-0.523,-0.671,P〈0.05),而196a2rs11614913位点CT和非小细胞肺癌易感性存在正向相关性(r=0.692,P〈0.05)。结论 miRNAs 149rs2292832位点CG和196a2rs11614913位点CC、CT和非小细胞肺癌均存在一定相关性,且和病情严重程度也存在一定关系,可将其作为非小细胞肺癌诊断和治疗的重要依据。
Objective To explore the association of miRNAs gene polymorphisms with susceptibility of nonsmall cell lung cancer.Methods Fifty new cases of patients with non-small cell lung cancer from January 2012 to June 2014 were selected as the study group,and another 50 cases of healthy volunteers for the normal physical examination in our hospital outpatient service center during the same period were selected as the control group.The miRNAs gene polymorphisms of blood of the research subjects between the two groups were compared,between which the clinical relationships with the susceptibility of non-small cell lung cancer were analyzed.Results The constituent ratios of 149rs2292832 miRNAs locus CG and 196a2rs11614913 locus CC in the study group were significantly lower than those of the control group(P〈0.05),and the constituent ratio of 196a2rs11614913 locus CT in the study group was significantly higher than that of the control group(P〈0.05).There were significant differences in 149rs2292832 miRNAs locus CG and 196a2rs11614913 locus CT and CC among the different samples in the study group(P〈0.05),and there were negative correlations between the 149rs2292832 locus CG and196a2rs11614913 locus CC with the susceptibility of non-small cell lung cancer(r =-0.523,-0.671,P〈0.05),while there was a positive correlation between 196a2rs11614913 locus CT with the susceptibility of nonsmall cell lung cancer(r = 0.692,P〈0.05).Conclusion There are certain correlations between the miRNAs gene polymorphisms with susceptibility of non-small cell lung cancer,as well as the severity of illness,which can be used as important basis for the diagnosis and treatment of non-small cell lung cancer.
