10-去乙酰基紫杉醇配伍紫杉醇对两种肿瘤细胞 生长抑制的作用

目的:观察紫杉醇配伍紫杉烷类成分10-去乙酰基紫杉醇(10-DAT)对A549、LS174T肿瘤细胞株的体外抑制作用.方法:采用A549、LS174T肿瘤细胞株进行MTT细胞毒性实验,观察10-DAT单独给药、10-DAT配伍紫杉醇对两种肿瘤细胞的生长抑制作用.结果:10-DAT单独使用时对LS174T细胞有一定的生长抑制作用,对A549则无抑制生长作用;但配伍紫杉醇反而会拮抗紫杉醇对A549和LS174T细胞的毒性作用.结论:10-DAT单独使用对LS174T细胞具有生长抑制作用,在高浓度下会拮抗紫杉醇对LS174T和A549细胞的细胞毒性,其原因可能源自两个化合物对微管蛋白的竞争性结合,确切机制有待进一步深入研究.

英文摘要:

Objective: To observe the growth inhibitory effect of paclitaxel combined with 10-DAT which is a kind of taxanes components on A549 and LS174T tumor cell lines in vitro. Methods: MTT cytotoxity assay was conducted with A549 and LS174T tumor cell lines to observe the growth inhibitory effect of 10-DAT administered alone and 10-DAT combined with paclitaxel on the two kinds of tumor cells in vitro. Results: When 10-DAT was administered alone, the growth of LS 174T cells had been inhibited in a concentration-dependent manner but no effect on A549; however, the compatibility with paclitaxel could antagonize the toxicity of paclitaxel to A549 and LS 174T cells instead. Conclusion: 10-DAT administered alone has growth inhibitory effect on LS 174T cells, but the toxicity of paclitaxel to A549 and LS 174T cells can be antagonized in heavy concentration, which may be associated with the competitive integration of the two compounds to tubulin protein and the exact mechanism needs further study.
