将实验室保藏的5株根瘤菌(S1、S2、S3、S6、S8)分别与解磷菌(P3)、硅酸盐细菌(K5)进行生物拮抗测试,结果表明,S2、S3、S6可与P3、K5进行混合培养。选择出与P3、K5混合培养促进解磷能力良好的根瘤菌株S6,解磷量为6.51mg.L-1。进一步进行混合培养培养基优化,优化后的最佳碳源为蔗糖,解磷量为9.81mg.L-1;最佳氮源为蛋白胨,解磷量为7.94mg.L-1。通过混合培养发酵条件的筛选,由正交试验得出最优发酵条件为:初始pH值为7.0;接种比例为S6:P3:K5=1:1:3;接种顺序P3→K5S6,其间相隔6h,此时解磷能力最强。
Laboratory preserved strains,Rhizobium(S1,S2,S3,S6,S8),phosphate solubilizing bacteria(P3) and silicate bacteria(K5) were mixed culture.Biological antagonism test result showed that S2,S3,S6 can mix culture with P3 and K5,a well-mixed culture rhizobial strains was selected as S6,Phosphorus concentration was 6.51mg·L-1.The optimized best carbon source was sucrose,phosphate-solubilizing capacity of 9.81mg·L-1;best nitrogen source was peptone,phosphate-solubilizing capacity of 7.94mg·L-1.Optimization of fermentation conditions: pH=7;Inoculation ratio S6:P3:K5=1:1:3;Inoculation order is in 6 hours with P3→K5S6.
