目的观察氟化钠对体外培养小鼠睾丸间质细胞凋亡的影响,以及对睾丸间质细胞中Bcl-2和Bax表达的改变。方法体外培养的小鼠睾丸间质细胞,将细胞分4组,加入不同浓度的氟化钠(0、5、10和20 mg/L),染毒0、24、48、72、96和120 h后采用MTT法检测各组细胞的增殖,并用免疫组织化学法检测氟化钠对小鼠睾丸间质细胞Bcl-2和Bax的表达。结果氟化钠作用24和48 h后,20 mg/L氟化钠对睾丸间质细胞增殖率有明显抑制作用,而5和10 mg/L组没有明显影响;10 mg/L氟作用72 h后,明显抑制睾丸间质细胞的增殖;各浓度氟作用96和120 h后均表现明显抑制作用。与对照组比较,染氟组睾丸间质细胞凋亡显著增加(P〈0.01),Bax表达增加(P〈0.01),而Bcl-2表达减少(P〈0.01)。结论氟化钠对小鼠睾丸间质细胞的增殖有抑制作用,并诱导小鼠睾丸间质细胞凋亡。氟化钠影响体外培养小鼠间质细胞凋亡及Bcl-2和Bax表达,证实了氟化钠对小鼠间质细胞的发育有毒性作用。
Objective To observe the effects of sodium fluoride on apoptosis and to detect the expressions of Bcl-2 and Bax in mouse leydig cells.Methods Leydig cells cultured in vitro were divided into 4 groups which were in DMEM/F12 medium respectively containing 0,5,10,20mg/L sodium fluoride.After each group was treated for 0,24,48,72,96,120 h,cell proliferation in each group was detected by MTT.The expressions of Bcl-2 and Bax were detected by immunohistochemistry.Results After the cells were treated for 24 h and 48 h,sodium fluorid(20mg/L) significantly inhibited the proliferation of Leydig cells,but there was no significant differences between 5 mg/L and 10 mg/L groups.After the cells were treated for 72 h,sodium fluorid(10mg/L) significantly inhibited the proliferation of leydig cells.After the cells were treated for 96 h and 120 h,sodium fluorid(5 mg/L,10 mg/L,20 mg/L) significantly inhibited leydig cells proliferation.Compared with control group,the apoptotic and the expressions of Bax in leydig cells increased(P〈0.01) in treatment groups.Conclusion Sodium fluorid can efficiently inhibit the proliferation,and promote the apoptosis of leydig cells.The results suggest that sodium fluoride could promote leydig cell apoptosis and increase the expressions of Bcl-2 and Bax in vitro,and it was also confirmed the cytotoxic effect of sodium fluorid in mouse leydig cells.
