[目的]分析亚东鲑线粒体COI与COⅡ基因遗传多样性。[方法]分别从西藏亚东河与亚东鲑养殖站采集野生与养殖亚东鲑各15尾样品,对其线粒体DNACOI、COⅡ基因全序列进行PCR扩增、测序比对。[结果]序列长度分别为631、634bp。序列分析结果显示:30条亚东鲑线粒体DNACOI、COⅡ序列分别检测到1种、3种单倍型,均无碱基插入、缺失,COⅡ序列有2个位点出现碱基替换;COI、COil序列T、C、A和G碱基平均含量分别为29.5%、28.97%,30.6%、28.47%,22.5%、27.03%和17.4%、15.53%,其中A+T的含量(52.O%、56.0%)均显著高于G+c含量(48.O%、44.0%),均表现出明显的碱基偏倚;COI、COⅡ单倍型多态性(h)与核苷酸多态性("iT)值分别为0、0.80与0、0.0001;根据Kimura双参数模型构建基于线粒体COI、COU序列的系统进化树,两亚东鲑鱼遗传距离分别为0、0.0024。【结论】西藏亚东鲑的遗传多样性水平较低,且养殖和野生群体无遗传分化。
[ Objective ] The aim was to analyze genetic diversity analysis of Salmo truttafario based on CO I and CO lI. [ Method ] Toal of 30 wild and cultured Salmo truttafario were collected from Tibet Yadong River and the nursery, respectively (15 each). The PCR amplification and sequencing were done. [ Result] The mitochondrial DNA CO I and CO lI gene sequence of Salmo truttafario was gotten, and the length were 631 and 634 bp, respectively. Sequence analysis results showed that 30 Salmo truttafario mitochondfial DNA CO I and CO H sequences contented 1 and 3 three kinds of haploid type, respectively. Neither of them had base insertions and deletions, CO II sequence had two loci base substitution. Average content of T, C, A and G bases of CO | and CO lI sequence were 29.5% and 29.5% , 30.6% and 28.47%, 22.5% and 27.03%, 22.5% and 27.03%, and the content of A + T (52.0% and 56.0% ) were significantly higher than that of G + C content (48.0% and 44.0% ), which showed a significant base bias. Haploid type polymorphism (h) and nucleotide polymorphism (π) val- ue of CO I and CO II were 0 and 0.80, 0 and 0.000 1 ,respectively. According to Kimura double parameter model building, system evolution- ary tree based on mitochondrial CO I and CO II sequence showed that genetic distance of two population were 0 and 0. 002 4, respectively. [ Conclusion i All results suggested that the genetic diversity of Salmo truttafario in Tibetan was not obvious, and there was no genetic differenti- ation between the cultured and wild population.
