目的观察香丹注射液的抗血管收缩效应,并且探讨其作用机制。方法利用体外血管条张力测定技术,分别用1g/L(低剂量香丹组)及10g/L(高剂量香丹组)香丹注射液预孵育大鼠胸主动脉体外血管模型20min,并与对照组(以0.9%氯化钠溶液孵育20rain)比较,观察香丹注射液的抗血管收缩功能。以棉签去除32条血管环的内皮细胞(E^-组),观察10g/L香丹注射液对E组的抗收缩效应,并与未去除内皮细胞组(E^+组)进行比较。分别以四乙胺(TEA)10mmol/L(TEA组)或磷酸盐缓冲液(PBS组)预作用20rain,再加入10g/L香丹注射液预孵育血管环20min后,观察并比较血管环的收缩反应率。结果高剂量香丹组与低剂量香丹组间血管环对0.01、0.1、1、10μmol/L去甲肾上腺素(PE)的收缩反应率的差异均无统计学意义(P值均〉O.05),但均显著高于对照组(P值均〈O.05)。10g/L香丹注射液处理后,E^+组与E^-组间血管环对O.01、0.1、1、10μmol/LPE的收缩反应率的差异均无统计学意义(P值均〉0.05),但均显著低于对照组(P值均〈0.05)。TEA组的血管环对0.01、0.1、1、10μmol/LPE的收缩反应率均显著高于PBS组(P值均〈O.05)。结论复方香丹注射液具有抗血管收缩的效应,这一作用呈非内皮依赖性,与开放钾离子通道有关。
Objective To investigate the effects of Xiangdan injection against vasoconstriction and its mechanisms. Methods In this study, 1 g/L Xiangdan injection (low dose group) and 10 g/L Xiangdan injection (high dose group) were applied in thoracic aorta of rats in vitro. 0.9% sodium chloride solution was taken as control. Vascular tension assay was used to determine the effect of Xiangdan injection against vasoconstriction. Endothelium of 32 vascular rings were removed with a cotton swab (E- group). The effect of 10 g/L Xiangdan injection against contraction in the E- group was observed and compared with the E+ group (normal endothelial cells). Ethylamine (TEA, 10 mmol/L) was used to pretreat the vascular rings for 20 min, then 10 g/mL Xiangclan injection was added for another 20 min incubation. The contraction of vascular rings was observed finally. Results There were no significant difference in the contractile responses of vascular rings to different concentrations of norepinephrine (0. 01, 0. 1, 1, and 10 μmol/L) between high-and low-dose Xiangdan injection pretreatment groups (all P〉0.05), but the responses in the two groups were significantly higher than control group (all P〈 0.05). There were no significant difference in the contractile responses of vascular rings to different concentrations of norepinephrine (0.01, 0. 1, 1, and 10 μmol/L) between E- group and E+ group (all P〉0.05), but the responses in the two groups were significantly lower than that in control group (all P〈0.05). The contractile responses of vascular rings to different concentrations of norepinephrine (0.01, 0. 1, 1, and 10 μmol/L) in TEA group were significantly higher than that in phosphate buffered solution (PBS) group (all P〈0.05). Conclusion Xiangdan injection has anti-vasoconstriction effect. This response is endothelium-independent and might be related with the opening of potassium channel. (Shanghai Med J, 2012, 35: 692-694)
