目的探讨振动与黄芪结合对后肢悬吊14 d引起的大鼠比目鱼肌萎缩的对抗效应。方法 30只雌性SD大鼠按体重匹配原则随机分为同步对照(Con),后肢悬吊(HU)和悬吊+治疗(HU-V-R)3组,每组10只。HU-V-R组大鼠在悬吊14 d期间,在大鼠的右下肢比目鱼肌肌腹部位施加"短时多次的间歇式"高频正弦波振动(100 Hz,0.5 mm);同时,给予每日1次的黄芪注射液腹腔注射(5 mL/kg)。14 d后,取比目鱼肌称量其湿重,计算湿重/体重;并进行比目鱼肌mATPase组织化学染色。结果与对照组比较,HU组大鼠在悬吊14 d后,比目鱼肌湿重、湿重/体重以及肌纤维横截面积均明显减小,同时Ⅰ型肌纤维比例减小而ⅡA型肌纤维比例增加(均P〈0.05)。与HU组相比,HU-V-R组大鼠比目鱼肌的湿重、湿重/体重以及肌纤维横截面积明显增加,同时Ⅰ型肌纤维所占比例也明显增高(均P〈0.05)。结论振动与黄芪结合能有效地对抗后肢悬吊引起的大鼠比目鱼肌萎缩。
Objective To investigate whether muscle vibration combined with Radix Astragali could prevent soleus muscle atrophy during 14-day hindlimb unloading(HU).Methods Thirty mature female Sprague-Dawley rats were randomly assigned to ambulatory control(Con),HU alone(HU),and HU submitted to Radix Astragali peritoneal injection associated with daily intermittent muscle vibration(HU-V-R)groups.Throughout the 14-day HU period,vibration(100 Hz;0.5 mm)was applied to the unloaded soleus muscle,and Radix Astragali parenteral solution peritoneal injection(5 mL/kg;once per day)was exerted in rats of HU-R-V group.Soleus muscles were examined 14 days later.The wet weight of the soleus muscles and the ratio of wet weight to body weight were measured.The soleus muscles were cut perpendicularly to its longitudinal axis into serial 10 μm thick cross-sections using a cryostat microtome at-20℃.Serial cross-sections were treated with adenosinetriphosphatase(ATPase)stain.Results In HU groups,as compared with the control groups,atrophy was characterized by a significant decrease in the soleus muscle wet weight,the ratio of wet weight to body weight,and in the fiber cross-sectional area.As well as there was a decrease in type Ⅰ fibers and an increase in ⅡA fibers(all P0.05).Application of vibration combined with Radix Astragali during HU significantly attenuated the atrophic response(all P0.05).Moreover,The relative proportion of type Ⅰ fibers was significantly recovered in HU-R-V groups(all P0.05).Conclusion Vibration combined with Radix Astragali is an effective countermeasure for muscle atrophy induced by HU.
