目的 探讨甘草酸二铵(DG)对胰腺癌Bx PC-3细胞株的影响。方法 胰腺癌Bx PC-3细胞株随机分成对照组,5-氟尿嘧啶(5-FU)组(50μg/ml),DG高、低剂量组(48,24μg/ml)。采用四甲基偶氮唑蓝比色法(MTT)检测胰腺癌Bx PC-3细胞株的增殖抑制率;采用细胞划痕法检测胰腺癌Bx PC-3细胞株的运动能力;电子显微镜观察胰腺癌Bx PC-3细胞株镜下形态结构;采用免疫组化和RT-PCR检测胰腺癌Bx PC-3细胞株中Raf-1水平。结果 与对照组比较,5-FU组及DG高、低剂量组细胞OD值明显降低(P〈0.05),运动能力明显降低(P〈0.05),胰腺癌Bx PC-3细胞株形态结构明显损伤,Raf-1水平明显降低(P〈0.05)。结论 DG通过抑制Raf-1在胰腺癌治疗中发挥重要作用。
Objective To study the effects of diammonium glycyrrhizinate(DG) on BxPC-3 cell line of pancreatic cancer. Methods BxPC-3 cell lines of pancreatic cancer were randomly divided into control ,5-Fluorouracil( 5'-FU, 50 μg/ml), DG high-dose (48 μg/ml ), DG low-dose (24 μg/ml) groups. The MTT was used to determine OD values of BxPC-3 cell line of pancreatic cancer. The cell scratch was used to determine the exercise capacity of BxPC-3 cell line of pancreatic cancer. Morphology of BxPC-3 cell line of pancreatic cancer was ob- served by optical electron microscopy. The immunohistochemistry and real-time-PCR were applied for detecting rapidly accelerated fibrosareo- ma(Raf) -1 levels in BxPC-3 cell line of pancreatic cancer. Results Compared with that of control group, OD values of BxPC-3 cell line of pancreatic cancer in 5-FU, DG high-dose and DG low-dose groups were significantly decreased( P〈0.05 ), exercise capacity were decreased ( P〈0.05 ). Compared with those of control group, BxPC-3 cell line of DG high-close and medial-dose groups were damaged normal morpho- logical structure, Raf-1 level was significantly decreased( P〈0.05 ). Conclusions DG plays a certain role in the treatment of BxPC-3 cell line of pancreatic cancer through inhibiting Raf-1.
