目的利用静电纺丝法制备载有环丙沙星的聚羟基丁酸酯电纺缓释纤维毡。方法利用扫描电镜考察了聚合物相对分子质量、聚合物溶液的浓度、电压及载药量对于纤维毡形态的影响。通过平板法和比浊法测定评价环丙沙星-聚羟基丁酸酯(CPX-PHB)电纺纤维毡的体外抑菌效果。结果增大所施加电压、PHB溶液浓度、PHB相对分子质量及载入药物CPX有利于电纺出具有直径小于2μm的均匀纤维的纤维毡。CPX-PHB电纺纤维毡对于金黄色葡萄球菌的生长在静止和动态环境中均具有明显的抑制效果。结论静电纺丝技术是一种简便的制备方法,利用其制备的CPX-PHB电纺纤维毡在防止手术后粘连和感染方面具有潜在的应用前景。
OBJECTIVE To prepare eelectrospun polyhydroxylbutyrate fibrous mats containing ciprofloxacin by electrospinning. METHODS The surface morphology of the electrospun fibrous mats was investigated by scanning electron microscope. In vitro bacteriostasis function was investigated with the determination of inhibition zone diameters on agar plate and the bacterial inhibition percentage by the absorbance at 625 nm in UV-VIS spectroscopy. RESULTS The uniform fibers without beads were acquired by increasing the applied voltage, poly-hydroxylbutyrate concentration, molecular weight and drug concentration. The CPX-PHB fibrous mats were effective to inhibit S. aureus growth in both a static (agar) and a dynamic (liquid) enviroment. CONCLUSION It is clear that the CPX-PHB electrospun scaffolds hold a future potential to prevent post-surgical adhesions and infections. And electrospinning is a simple and stable method to prepare CPX-PHB electrospun fibrous mats.
