目的:探讨短期和长期运动预适应对大鼠血液和心脏降钙素基因相关肽(calcitonin gene-relatedpeptide,CGRP)的影响及其对心肌保护的作用机制;方法:健康雄性SD大鼠75只,随机分为对照组(C组)、3天运动预适应组(3dEP组)和3周运动预适应组(3周EP组)。3dEP组和3周EP组分别进行3天和3周的间歇跑台训练建立短期和长期运动预适应动物模型。用酶联免疫法检测血清CGRP含量的变化,用western blot方法和免疫组织化学方法检测心脏CGRP定量和定位表达的变化;结果:与C组比较,3dEP组血清CGRP含量变化不明显,3周EP组血清CGRP含量明显升高(P〈0.05);与C组比较,3dEP组心脏CGRP表达明显减少,CGRP免疫反应阳性表达面积和平均光密度明显降低(P〈0.05),3周EP组心脏CGRP表达变化不明显,CGRP免疫反应阳性表达面积和平均光密度无明显变化;结论:短期运动预适应使心脏CGRP表达下降,提示短期运动预适应可促进心脏CGRP的释放,对心肌产生保护作用;长期运动预适应使血清CGRP水平升高,提示长期运动预适应可提高机体CGRP的释放水平,对心肌产生保护作用。
Objective:The effects of short-term and long-term exercise preconditioning on calcitonin gene-related peptide (CGRP) in the blood and heart of rats and the mechanism of cardioprotection were studied.Method:Seventy five healthy male SD rats were randomly divided into control group (C),3 days (3 d EP) and 3 weeks (3 w EP) exercise preconditioning groups.3 d EP and 3 w EP groups performed 3 days and 3 weeks interval treadmill exercise respectively in order to establish short-term and long-term exercise preconditioning animal models.The content of CGRP in serum was detected with enzyme immunoassay method.The expression of CGRP in heart was showed by western blot and immunohistochemistry.Result:As compared with the group C:the content of serum CGRP changed insignificantly in group 3 d EP,the content of serum CGRP increased significantly in group 3 w EP (P0.05); the expression of cardiac CGRP decreased significantly in group 3 d EP,the positive area and optical density of CGRP immunoreaction were significant lower in group 3 d EP (P0.05),the expression of cardiac CGRP changed insignificantly in group 3 w EP,the positive area and optical density of CGRP immunoreaction changed insignificantly in group 3 w EP.Conclusion:Short-term exercise preconditioning can decrease the expression of CGRP in heart,which demonstrated myocardial protection is mainly associated with the increase of CGRP released in heart.Long-term exercise preconditioning can improve the level of CGRP in serum,which demonstrated myocardial protection is mainly associated with the increase of CGRP released in body.
