As a class of important natural product, many steroidal saponins were proved to own promising biological property, such as antitumor activity. The syntheses, structural modifications and bioactivity investigations of this class of compounds have led to some primary structure-activity relationships, which are of great meaning for the further development of novel antitumor agents. The progress on the chemical synthesis of steroidal saponins with antitumor activity in the past 10 years was reviewed.
