目的:探讨六味地黄颗粒在防治支气管哮喘过程中的抗炎机制及其作用靶位。方法:将健康雄性SPF级SD大鼠1607,随机分为8组,即六味地黄颗粒组(A组)、六味地黄颗粒加地塞米松组(B组)、地塞米松组(C组)、六味地黄加布地奈德组(D组)、布地奈德组(E组)、模型组(F组)、生理盐水对照组(G组)及正常对照组(H组),每组20只。第1、8天除C、H组外,其余各组均以卵蛋白(OVA)辅以氢氧化铝注射致敏,G组以生理盐水腹腔注射代替;2周后除G、H组外,其余各组雾化吸入卵蛋白以激发建立哮喘模型,G组雾化吸入生理盐水代替.正常对照组不予任何处理。从实验第8天开始,除F、G、H组外,其余各组分别用药干预,G组及F组用生理盐水灌胃代替,H组不予处理,造模时间共28天。取材后采用ELISA法分别检测血清中NO浓度及肺组织匀浆液中NOS浓度。结果:在实验大鼠肺组织匀浆液NOS含量中,A、B、D组含量相对于F组明显偏低、且其差异存在统计学意义(尸〈0.05),在实验大鼠血清NO含量中,与F组比较:A、B组大鼠血清中NO的含量无明显偏低,差异不具有统计学意义(P〉0.05);C、D组大鼠血清NO含量均明显偏低,差异具有统计学意义(P〈0.05)。结论:抑制哮喘肺组织NOS的合成与释放可能是六味地黄颗粒用于治疗哮喘并减轻气道炎症的重要机制之一。
Objective: To observe the anti-inflammatory mahine processed and effective locus of LiuWeiDiHuang Granula on the process of prevention and cure asthma. Methods: 160 healthy male SD rats were randomly divided into eight groups :LiuWeiDiHuang Granula group (group A), LiuWeiDiHuang Granula and hexadecadrol group (group B ), hexadecadrol group (group C ), LiuWeiDiHuang Granula and budesonide group (group D ), budesonide group (group E ), asthma model group (group F), nagtive control group (group G) and normal control group (H group). 20 rats were included in each group .On the first and eighth day, all the other 6 groups were sensitized with ovalbumin (OVA) combined aluminum hydroxide as adjuvant by injecting into the abdominal cavity except group H, while group G was sensitized with physiological saline injecting instead. Two weeks later, the other 6 groups were followed by aerosolized OVA to excite the asthma models while group G was followed by aerosolized physiological saline and group H without any deal. From the eighth day of the experiment, rats were accepted the predetermined medicine intervention in medicine administration groups while group G and group F was given intragastric administration with physiological saline and group H without any deal. All the copy process of animal model was 28 days. Results: the NOS content in Lung tissue homogenate liquid in group A, group B and group D were lower than that in group F (P〈0.05). The blood serum NO difference was statistically significance(P〈O.05). Conclusion: LiuWeiDiHuang Granula may reduce airway inflammation and cure asthma by restrain the synthesis and release of NOS in lung tissue as a main mechanism.
