目的:研究胰腺脱细胞支架三维(3D)培养对小鼠β细胞系MIN6细胞生长及功能的影响。方法灌注法制备胰腺脱细胞支架,循环灌注培养法在胰腺脱细胞支架中3D培养小鼠MIN6细胞,检测其功能;其结果与2D平面培养比较。将含细胞的胰腺支架移植入糖尿病小鼠背部皮下并检测其功能。结果胰腺脱细胞支架检测未见细胞结构,细胞外基质成分保存完好,血管结构保存完整。支架内剩余DNA定量检测结果为(40.7±2.1)ng/mg,体内种植显示其生物相容性良好。胰岛素基因表达的检测显示,MIN6细胞在胰腺脱细胞支架3D培养后的功能发挥优于2D平面培养(P<0.05),移植后能够较好的控制糖尿病小鼠的血糖水平。结论MIN6细胞在胰腺脱细胞支架3D培养体系中定植及功能发挥优于传统2D平面培养。
Objective To study the effects of three‐dimensional(3D) culture in decellularized pancreatic scaffold on the regeneration and function of the murineβcells(MIN6 cells) .Methods The decellularized pancreatic scaffold was manufactured by perfusion .MIN6 cells were cultured in the decellularized pancreatic scaffold by continuous perfusion of culture medium .The functions of MIN6 cells cultured in the decellularized pancreatic scaffold(group A) and on the culture dish(group B) were tested and compared .The recellularized pancreatic scaffold was implanted subcutaneously on the dorsal side of the diabetic mice to evaluate its function in vivo .Results There was no cellular composition left in the decellularized pancreatic scaffold ,while the extracellular matrix and whole vasculature were reserved .The quantification of the remaining DNA in the scaffold was (40.7 ± 2.1) ng/mg .In vivo implantation demonstrated that the biocompatibility of the decellularized pancreatic scaffold was well . The function of MIN6 cells in group A was better than that in group B and there was statistical significance in the expression of insulin gene between two groups(P〈0 .05) .The blood glucose level of the diabetic mice was well controlled after transplantation of the recellularized pancreatic scaffold . Conclusion The regeneration and function of the MIN6 cells cultured in the 3D decellularized pancreatic scaffold are better than those cultured on traditional culture dish .
