目的 考察厚朴汁炙远志含药血清对小鼠胃Cajal间质细胞内离子活性来影响胃动力的细胞钙通道机制.方法 采用体外小鼠胃Cajal间质细胞和含药血清共孵.实验分成:空白组、厚朴组、远志组、远志配厚朴组、厚朴汁炙远志含药血清高、中和低剂量组.采用分光光度法检测细胞内外钙离子浓度和微量ATP酶、ELISA法检测1,4,5-三磷酸肌醇(IP3)浓度.结果 远志组的胞内游离Ca2浓度、ATP酶活性、和IP3浓度与空白组比较均显著升高(P<0.05,P<0.01),而厚朴组、远志配厚朴组,厚朴汁炙远志含药血清高和中剂量组的Ca2浓度、ATP酶活性、IP3浓度与远志组比较,显著降低(P <0.05,P<0.01).结论 远志能抑制胃Cajal间质细胞活性,使细胞钙离子内流,产生自由基导致细胞成分破坏,诱发细胞凋亡;而厚朴汁炙远志能缓解细胞钙超载,维持细胞稳态,从而缓解胃动力障碍.
AIM To study Polygalae Radix processed with the decoction of Magnoliae officinalis Cortex on cal- cium ion channel of mouse gastric interstitial cells of Cajal (ICCs) and their gastric dynamics. METHODS ICCs were co-cultured in vitro with serum containing herbs and were divided into seven groups: blank, Magndiae offic- inulis Cotex, Polygalae Radix, Pologalae Radix plus Magnoline officinalis Cortex, high, middle and low dose of Polygalae Radix processed with the decoction of Magnoline officinalis Cortex group. Spectrophotometry was used to determine the intracellular calcium ion concentration and ATP enzyme. The concentration of IP3 was detected by ELISA method. RESULT Compared with the blank group, intracellular free Ca2+ concentration, ATP enzyme activity, and IP3 concentration in Polygalae Radix group were all significantly higher (P 〈0.05, P 〈0. 01 ). Whereas compared with Polygalae Radix group, ATP enzyme activity and IP3 concentration in Magndiae officinulis Cortex, Polygalae Radix plus Magnoliae officinalis Cortex, high and middle dose of Polygalae Radix processed with the decoction of Magnoline officinalis Cortex group decreased significantly ( P 〈 0. 05, P 〈 0. 01 ). CONCLU- SION Polygalae Radix can inhibit the activity of ICCs motility to make intracellular calcium inflow and produce free radicals to cause the damage to cell components and cell apoptosis. Polygalae Radix processed with the decoc- tion of Magnoline officinalis Cortex can alleviate the effect of calcium overload while maintain the cellular homeosta- sis of ICCs to relieve the gastric motility disorder.
