目的考察远志与厚朴配伍对胃肠组织一氧化氮(NO)、胃窦血管活性肠肽(VIP)的影响,探讨厚朴缓解远志胃排空抑制的作用机制。方法采用硝酸还原酶法,考察了远志、厚朴、远志与厚朴1:1、1:2、1:3组对大鼠胃肠组织NO含量的影响;采用免疫组织化学法,考察了给药组对大鼠胃窦肌层VIP的影响。结果远志能显著增加大鼠胃窦组织NO含量;与单味远志组比较,远志与厚朴配伍各组均能显著降低的胃窦组织NO含量,但对十二指肠组织NO含量无显著影响。单味远志能明显增强大鼠胃窦肌层的VIP免疫阳性产物表达;远志与厚朴配伍1:1组的VIP表达量明显低于单味远志。结论单味生远志导致胃排空障碍可能与其增强胃窦组织NO含量、增强该区域肌层VIP高表达有关;厚朴可能通过降低胃窦NO含量、下调胃窦区VIP表达发挥缓解其胃排空抑制作用的。
Objective To investigate the effects of the compatibility of Magnolia Officinalis Cortex and Polygalae Radix on nitrogen monoxidum(NO) of gastrointestinal tract tissues and vasoactive intestinal peptide(VIP) of stomach,and explore the mechanisms of Magnolia Officinalis Cortex on relieving the inhibitory action of gastric emptying that induced by Polygalae Radix.Methods The effects of Polygalae Radix,Magnolia Officinalis Cortex,and the compatibility of the two herbs(with the ratios of 1:1,1:2 and 1:3) on NO content of gastrointestinal tract tissues of rats were investigated by using nitrate reductase method.The effects of the above mentioned herbs on VIP content of muscular layer of sinus ventriculi of rats were investigated by using immunohistochemical method.Results Polygalae Radix could significantly increase NO content of sinus ventriculi of rats(P〈0.05).Compared to single herb of Polygalae Radix,all the combinations of the two herbs could significantly decrease NO content of sinus ventriculi of rats(P〈0.05),but showed no significant in?uence on NO content of dodecadactylon.Polygalae Radix could significantly enhance the immuno-positive expression of VIP of the muscular layer of sinus ventriculi of rats(P〈0.05),and the effect of the combination of Polygalae Radix and Magnolia Officinalis Cortex(with the ratio of 1:1) on VIP content was much lower than single herb of Polygalae Radix(P〈0.05).Conclusion The inhibitory action of gastric emptying that induced by Polygalae Radix might be associated with its effects of increasing NO content of gastrointestinal tract tissues and enhancing the positive expression of VIP of the muscular layer of sinus ventriculi.Meanwhile,Magnolia Officinalis Cortex could relieve the inhibitory action of gastric emptying by means of decreasing NO content of gastrointestinal tract tissues and reducing the positive expression of VIP of the muscular layer of sinus ventriculi.
