目的:探讨野生型p53对Wnt通路的调节以及对肺癌细胞生物学行为的影响。方法:分别利用野生型p53质粒及siRNA,分别转染和干扰A549细胞中的野生型p53,而后用Western blot检测A549细胞中β-catenin和cyclin D1的表达,用荧光素酶报告基因法检测Wnt通路活性,用MTT和Transwell实验检测A549细胞的增殖和侵袭能力。结果:转染野生型p53后能够明显下调A549细胞中β-catenin和cyclin D1表达(P〈0.01),抑制肺癌细胞的增殖和侵袭(P〈0.05),而干扰p53可以明显地上调β-catenin和cyclin D1表达(P〈0.01),增强肺癌细胞的增殖力和侵袭力(P〈0.05)。结论:野生型p53负向调控A549细胞中的Wnt信号转导通路,抑制其增殖和侵袭能力。
Objective:p53 is one of the most important tumor suppressors, the regulation of wild - type p53 on Wnt pathway in lung cancer cells is still not clear, we explore the wild type p53 function on Wnt pathway and lung cancer cell biology behaviors. Methods : Wild - type p53 plasmid and siRNA were used to transfect and knock down p53 in A549 cells,Western blot was used to detect β -catenin and cyclin D1 protein level, luciferase reporter gene was used to detect the activity of the Wnt pathway, MT/' and Transwell assay were used to detect the proliferation and invasion of A549 cells. Results: The transfection of wild - type p53 markedly reduced β - catenin and cyclin D1 in A549 cells ( P 〈0.01 ) and inhibited the proliferation and invasion (P 〈0.05 ) ,knock down of p53 up - regulated β - catenin and cyclin D1 ( P 〈 0.01 ), and enhanced proliferation and invasion in A549 cells ( P 〈 0.05 ). Conclusion: The wild -type p53 negatively regulates Wnt signaling transduction pathway and inhibits the proliferation and invasion of A549 cells.
