目的通过对谷胱甘肽(GSH)代谢研究来探索氟对成骨细胞氧化应激的影响。方法取材昆明小鼠乳鼠颅骨来源的成骨细胞进行原代培养并传至第3代,用高效液相色谱仪对不同浓度染氟的成骨细胞内GSH、氧化型谷胱甘肽(GSSG)含量进行检测。结果在含氟1.0、2.0mgF^-/L培养液中作用24-72h,成骨细胞的GSH水平多有不同程度的增高,在12.0、20.0mgF^-/L氟水平作用下,GSH水平达到最高;染氟24-72h的成骨细胞在0.5、1.0、2.0mgF^-/L培养液孵育下,细胞的GSSG水平有不同程度的下降(除48h的0.5、1.0mgF^-/L组细胞外);暴露于氟24h的成骨细胞在含氟0.5、2.0mgF^-/L培养液孵育下,细胞的GSH/GSSG比值有不同程度的提高;氟暴露48h组,成骨细胞在2.0、8.0mgF^-/L氟水平作用下,GSH/GSSG比值亦明显增加。结论低剂量氟作用下GSH含量和GSH/GSSG比值增加,而GSSG水平下降,说明成骨细胞处于活跃的抗氧化状态,进一步证明了成骨细胞内氧化应激水平明显增强。
Objective To study the effect of fluorides on oxidative stress by detecting the metabolism of GSH. Meth. otis Calvarial osteoblasts out of neonatal mouse were cultured and subcuhured to the third generation. Osteoblasts were exposed to varied concentration of fluoride for 24 h to 72 h in vitro and tested the level of GSH and GSSG by high performance liquid chromatographic ( HPLC ). Results The level of GSH obviously increased in osteoblasts exposed to 1,2 mgF^-/L for 24 h to 72 h, and the highest level was observed in the osteoblasts of 12,20 mgF^-/L group. The level of GSSG markedly decreased in osteoblasts exposed to 0. 5, 1,2 mgF^-/L for 24 h to 72 h ( except for 0. 5,1 mgF^-/L groups of 48 h ). The ratio of GSH/GSSG obviously increased in osteoblasts exposed to 0. 5, 2 mgF^-/L for 24 h and 2, 8 mgF^-/L for 48 h. Conclusions Low - dose fluoride enhanced the concentration of GSH and increased the ratio of GSH/GSSG, but reduced the concentration of GSSG, which suggested the active state of anti - oxidation, and further showed the increase of oxidative stress in osteoblasts.
