目的:研究钙拮抗剂(维拉帕米、地尔硫革和硝苯地平)对大鼠心肌缺血再灌注(I/R)革早期生长反应基因-1(Egr-1)表达的影响。方法:大鼠随机分为假手术组、I/R组、二甲基亚砜组、维拉帕米组、地尔硫革组、硝苯地平组。Westem blot法测定Egr-1蛋白表达水平的变化;RT-PCR法测定Egr-1 mRNA表达水平的变化。结果:与假手术组比,L/R组Egr-1蛋白及Egr-1 mRNA表达显著增高(P〈0.05);与I/R组比,维拉帕米组、地尔硫革组和硝苯地平组Egr-1蛋白及Egr-1 mRNA表达明显减少(P〈0.05)。结论:维拉帕米、地尔硫革和硝苯地平均可抑制缺血再灌注心肌Egr-1蛋白及Egr-1 mRNA的过度表达。
Objective: To study the effects of calcium antagonist(verapamil, diltiazem and nifedipine)on early growth response gene-1(Egr-1)expression in rats after myocardial ischemia-reperfusion(I/R). Methods: Rats were randomly assigned into Sham, I/R, DMSO, vempamil, diltiazem, nifedipine groups. The expression levels of Egr-1 protein and mRNA were examined by Western blot and RT-PCR, respectively. Results: Compared with sham group, Egr-1 protein and mRNA of I/R group overexpressed( P 〈 0.05). The expression levels of Egr-1 of vempamil, diltiazem, nifedipine groups significantly decreased, compared with I/R group( P 〈 0.05). Conclusion: Verapamil, diltiazem and nifedipine ean inhibit the overexpression of Egr-1 protein and mRNA in rots caused by I/R.
