采用正交设计法研究适合篦子三尖杉(Cephalotaxus oliveri)的ISSR—PCR反应体系,对影响ISSR—PCR的模板、引物、Mg^2+、dNTP、Taq酶等5个因素进行了优化。确定了篦子三尖杉ISSR—PCR扩增的最佳体系,即20μl的反应体系中包括:1μl 10×PCRbuffer、2.5mmol/L Mg^2+、0.2mmol/L dNTPs、0.25mol/L的ISSR引物、1.5UTaqDNA聚合酶、50ng模板DNA。运用优化的体系对江西武功山篦子三尖杉总DNA进行ISSR—PCR扩增,获得较好效果。
The orthogonal design was used to optimize ISSR - PCR amplification system, such as DNA template, primers, concentration of Mg^2+ , dNTPs and Taq DNA polymerase were investigated to optimized the ISSR -PCR reaction system of Cephalotaxus oliveri. A sutable ISSR - PCR reaction system was established, namely 20 lreaction system contaning: 1 μl 10 × PCRbuffer 2.5mmol/LMg^2+ , 0.2 mmol/L dNTPs, 0.25 μmol/L Primer, 1.5U TaqDNA polymerase, 50ngDNA. The optimized system laid the groundwork for ISSR molecular analysis of of Cephalotaxus oliveri.
